HEK293T cells were transfected with the pCMV6-ENTRY control (Cat# PS100001, Left lane) or pCMV6-ENTRY CRY1 (Cat# RC207152, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-CRY1 (Cat# TA809887)(1:2000). Positive lysates LY418241 (100ug) and LC418241 (20ug) can be purchased separately from OriGene.

  Equivalent amounts of cell lysates (10 ug per lane) of wild-type HeLa cells (WT, Cat# LC810HELA) and CRY1-Knockout HeLa cells (KO, Cat#LC834622) were separated by SDS-PAGE and immunoblotted with anti-CRY1 monoclonal antibody TA809887 (1:200). Then the blotted membrane was stripped and reprobed with anti-PCNA antibody as a loading control.

• Background: This gene encodes a flavin adenine dinucleotide-binding protein that is a key component of the circadian core oscillator complex, which regulates the circadian clock. This gene is upregulated by CLOCK/ARNTL heterodimers but then represses this upregulation in a feedback loop using PER/CRY heterodimers to interact with CLOCK/ARNTL. Polymorphisms in this gene have been associated with altered sleep patterns. The encoded protein is widely conserved across plants and animals.
